Gene Cloning Techniques

Description:
This workshop is aimed to provide basic understanding/knowledge of gene cloning and its applications in recombinant protein expression, purification and production. Major topics will include various cloning strategies, PCR, gene expression systems for production of recombinant proteins., recombinant plasmid purification/ restriction enzymes digestion analysis of constructs and expression/ purification of recombinant proteins.
Topics that will be covered:
The intensive workshop is specifically designed to address
• Gene cloning strategies &Plasmid construction
• Restriction digestion of pUC19 vector using EcoRI
• 5` End DNA modification of restriction digested plasmid sample [Addition of Poly Ts]
• DNA ligation
• Bacterial Transformation [using competent cells and cloned vector
• Blue white screening [checking for the transformed colonies].
• Recombinant plasmid and recombinant proteins puri¬fication.
• Transfection into mammalian cells.
Duration: 4 working days (3 hours/day).
Course structure: The course will constitute theoretical classes, demonstration and hand training.
Workshop Agenda:
1st day: Introduction in DNA recombinant technology, Restriction enzyme digestion, ligation of gene of interest and PCR amplification.
2nd day: Bacterial transformation.
3rd day: Purification of recombinant DNA and proteins.
4th day: Transfection of the recombinant DNA into mammalian cells.
